If i am creating a Server 2008 R2 VMware Template , do i need to make sure windows activation is done already and is sysprep required.

once the template is created i will use vmware customization wizard to deploy VM's using this template

You don't have to run sysprep, but you do need to have it available to
vSphere, which will use it when you deploy the template.
(which means when you turn it on for the first time, give it a couple
minutes, as the sysprep will cause a reboot, so you need to wait until
after it reboots before you log in the first time).
